NBSI ligands for the Clivia long Stocks shift fluorescent RNA
NBSI ligands are nonfluorescent in solution, but emit strong fluorescence upon forming tight complex with Clivia RNA aptamer. The fluorescent RNAs thus formed, Clivias, are the only long Stocks shift fluorescent RNA tags currently availabe for visualization of RNAs of various species in live cells and live animal.
NBSI fluorescence can be activated thousands fold by Clivia RNA aptamer. There are 14 kinds of NBSI ligands with different colors in NBSI series fluorescent dyes. They cover a wide range of emission wavelengths, named after the maximum emission wavelength. FR-Biotechnology currently offer NBSI580, NBSI574, NBSI595 and NBSI618, which have been well validated for mamalian cell studies.
- NBSI580 is the original NBSI ligand with brightest fluorescence.
- NBSI574 is recommended for BRET imaging when combined with nanoluc luciferase, which is useful for investigating RNA-protein interaction, or imaging RNA metabolism in vivo.
- NBSI595 together with HBC497 are validated for single excitation dual color imaging for RNAs tagged withClivia and Pepper, respectively.
- NBSI618 has longer emission wavelength and lower background fluorescence in cells.
- Customized size of above NBSI reagents and other NBSI ligands vairants are available on request.
Supplied as solid formulation. Reconstitute with DMSO (60 ul for S size, or 200 ul for L size as labeled on the vial) to make 1000X stock solutions.
We suggest users start with NBSI595, and HEK293T cells expressing Clivia or Clivia tandem arrays as positive control, before test your own cells and/or RNAs. As far as we know, cells of different origin expressing Clivia alone or Cliva array show bright fluorescence and low background when stained with NBSI595.
After the first success, users may proceed to test the performance of other NBSI ligands on your RNA, cells and instruments.
Important Notice: Customers should include at least one positive control plasmids (PET28C8MCS0 for E. coli cells or PAPU6C8MCS0 for mammalian cells) in their first order along with their choice of NBSI dyes. Alternatively, customers may order the Clivia Fluorescent RNA starter kits (Cat# PET28C8KIT or PAPU6C8KIT).
